Zymolyse and buffer, 2000 U is a combination of 1 mL buffer solution at pH 7.4 and 2000 U of lyophilized Zymolyase powder prepared from Arobacter luteus. It provides convenience to make fresh zymolyase stock solution at 2,000 U/mL without lossing enzymatic activities. It is used to effectively lyse cell walls of viable yeast cells by hydrolyzing glucose polymers at the ß-1,3-glucan linkages. The buffer contains 1 mL potassium phosphate with D-sorbitol at pH 7.4.
One lytic unit is defined as catalyzing a 10% decrease in absorbance at 800 nm at 30 °C in 30 minutes.
Assay condition: 50 mM potassium phosphate, pH 7.5, 10 mM 2-mercaptoethanol in 1 mL yeast cell suspension of Abs. 800 nm 0.8 to 1.0.
