SPINeasy DNA Kit for Soil is a high-performance soil gDNA extraction kit based on spin-column technology. It enables quick isolation of gDNA from soil in less than 30 minutes. Briefly, soil samples are placed into Lysing Matrix E tubes and used with FastPrep® Instruments from MP Biomedicals to effectively lyse various cells including bacteria, fungi, viruses, protists and more. A series of specially formulated reagents were developed to remove humic substances and other contaminants. Column S1 provided in the kit has high binding capacity and selectivity for gDNA. The combined components of the kit provide an indispensable tool for extracting high yields of pure gDNA from even the most difficult environmental samples. Extracted gDNA is ready for downstream analyses such as PCR, restriction digestion and sequencing.
SPINeasy DNA Kit for Soil is compatible with various soil types. Extracted gDNA is of high purity when undergoing spectrophotometric quantitation.
SPINeasy DNA Kit for Soil demonstrated better purity of extracted DNA when compared to the other competitor DNA kits. The purity of the extracted DNA was determined with spectrophotometric analysis by measuring and calculating the ratio of the absorbance at 260 nm and 280 nm (A260/280). Typically, A260/280 ratio ~ 1.8 is generally considered as high purity for DNA and A260/280 ratio ~ 2.0 is generally considered as high purity for RNA.
Remark 1: Purity 260/280: The genomic DNA extracted with SPINeasy DNA kit for Soil (in blue bar) was the only DNA kit among the comparator DNA kits to show high purity of DNA as assessed by its A260/280 ratio across all the soil sample types studied, which were about 1.80.
The level of contaminants in the DNA extracted was determined similarly with spectrophotometric analysis by measuring and calculating the ratio of absorbance at 260 nm and 230 nm. Strong absorbance around 230nm can indicate that organic compounds or chaotropic salts are present in the purified DNA. A ratio of 260nm to 230nm can help evaluate the level of salt carryover in the purified DNA. The lower the ratio, the greater the amount of thiocyanate salt is present, for example. The contaminants may come from the sample source such as humic acid and from the extraction procedure such as guanidine thiocyanate (GTC). To be generally considered as good purity of nuclei acids and free from the contaminants, the A260/280 ratio should be a 1.0 or above 1.0.
Remark 2: Purity 260/230: The genomic DNA extracted with SPINeasy DNA kit was generally free from the contaminants as assessed by its A260/280which were above 1.0 across all the soil sample types studied.
SPINeasy DNA Kit for Soil has consistently achieved better yield with good purity as compared to competitor kits. Total yield is obtained by multiplying the DNA concentration by the final total purified sample volume without visible shearing or smearing during gel electrophoresis.
