PRODUCT DESCRIPTION

YPD (YEPD) is a nutritionally rich medium for the growth of S. cerevisiae, containing only, peptone and yeast extract, without dextrose. This is useful for variations of YPD requiring alternate carbon sources. Glycerol can be added as a non-fermentable carbon source to make YEP-glycerol (YEPG) for the study of respiratory deficient mutants. Yeast extract and peptone provide carbon, nitrogen, minerals, vitamins, trace ingredients and other essential growth nutrients which stimulate the growth of yeasts. Application Notes YED Broth is used for routine growth of Saccharomyces cerevisiae for molecular biology studies. It is used as a base for making a variation of YPD with an alternate carbon source. Preparation Method : Media Preparation: Reagents are tested and chosen for their ability to promote optimum growth of S. cerevisiae when combined in YEP Broth formulations. Chosen reagents are combined in the proper proportions according to the YEP formulation and thoroughly blended for a uniform distribution. A sample of the blended formulation is used to prepare agar medium. The combinations are mixed to dissolve components and then autoclaved at 121 °C for 15 minutes.
Solubility : Soluble in Water(30 g/L).
Sterilization of Solutions : Combinations are mixed to dissolve components and then autoclaved at 121 °C for 15 minutes. Note: Autoclaving at higher temperatures or for longer periods will result in darkening of the sterile medium, (a darker color does not normally affect growth of cells, only the appearance of the sterile medium).
Typical Working Concentration : Effective concentration is 1 to 100 µM.
